Sitapin-Met Tablet 50/500mg

Sitapin-Met Tablet 50/500mg is an oral fixed-dose combination antidiabetic medication containing sitagliptin (50mg) and metformin hydrochloride (500mg). It operates through dual complementary mechanisms to optimize glycemic control in adults with type 2 diabetes. Sitagliptin, a competitive dipeptidyl peptidase-4 (DPP-4) inhibitor, preserves active incretin hormones (GLP-1 and GIP) by preventing their enzymatic breakdown, thereby stimulating glucose-dependent insulin secretion from pancreatic beta cells while suppressing postprandial glucagon secretion from alpha cells. Concurrently, metformin, a biguanide, acts primarily by activating AMP-activated protein kinase (AMPK) to suppress hepatic gluconeogenesis, decrease intestinal absorption of glucose, and improve peripheral insulin sensitivity and glucose uptake in skeletal muscle.Clinically, Sitapin-Met 50/500mg is indicated as an adjunct to diet and exercise to improve glycemic control in adult patients with type 2 diabetes mellitus when treatment with both sitagliptin and metformin is appropriate.The standard starting dose is 1 tablet taken orally twice daily with meals to reduce gastrointestinal side effects associated with metformin. Tablets should be swallowed whole with water and never crushed or chewed.It is contraindicated in severe renal impairment , acute or chronic metabolic acidosis (including diabetic ketoacidosis), and known hypersensitivity to sitagliptin or metformin. Common side effects include diarrhea, nausea, flatulence, abdominal discomfort, headache, and hypoglycemia when combined with insulin or sulfonylureas
