Sitapin-Met Tablet 50/1000mg

Sitapin-Met Tablet 50/1000mg is an oral fixed-dose combination antidiabetic medication containing sitagliptin (50mg) and metformin hydrochloride (1000mg). It works through two complementary mechanisms to optimize blood glucose regulation. Sitagliptin, a competitive dipeptidyl peptidase-4 (DPP-4) inhibitor, prevents the degradation of active incretin hormones (GLP-1 and GIP), thereby stimulating glucose-dependent insulin secretion from pancreatic beta cells while suppressing postprandial glucagon release from alpha cells. Concurrently, metformin, a biguanide, acts via AMP-activated protein kinase (AMPK) activation to inhibit hepatic gluconeogenesis, decrease intestinal glucose absorption, and enhance peripheral tissue sensitivity to insulin, increasing glucose uptake in skeletal muscles.Clinically, Sitapin-Met 50/1000mg is indicated as an adjunct to diet and exercise to improve glycemic control in adult patients with type 2 diabetes mellitus when higher metformin dosing is required alongside DPP-4 inhibition.The standard adult starting dose is 1 tablet taken orally twice daily with meals to minimize gastrointestinal adverse effects. Tablets must be swallowed whole with water and should not be crushed, split, or chewed.It is contraindicated in severe renal impairment , acute or chronic metabolic acidosis (including diabetic ketoacidosis), and known hypersensitivity. Common side effects include diarrhea, nausea, flatulence, abdominal pain, headache, and hypoglycemia when co-administered with insulin secretagogues
